![]() ![]() In this review we first consider the signalling pathways that mediate TNF responses, resulting in both eradication of infectious agents and, in some cases, concomitant tissue injury. Through its pro-inflammatory effects on various cell types, examples of activation, TNF plays a major role in the host defence to bacterial, viral and parasitic infections (Figure 1). Activation responses, defined here as reversible induction of specific gene expression that confers greater capacities to perform specific functions, typically lead to the recruitment and activation of leukocytes, producing inflammation. TNF is a pleiotropic cytokine that acts upon almost every differentiated cell type, eliciting a wide range of diverse cellular responses, including activation, cell death, survival, differentiation and proliferation.
